Shoot Induction, Multiplication, Rooting and Acclimatization of Black Turmeric (Curcuma caesia Roxb.): An Important and Endangered Curcuma Species

Curcuma caesia Roxb., commonly known as Kali Haldi or black turmeric, is one of the important species in genus Curcuma. This has been classified endangered due to drastic decrement this plant its natural habitat. C. overharvested for various purposes, including bioactive compound extraction fulfill pharmaceutical industry demand. Hence, study was conducted establish a protocol propagation via tissue culture techniques. In shoot induction stage, three basal medium formulations, Murashige and Skoog (MS medium), combination macronutrients B5 micronutrients (MSB5 medium) woody (WPM supplemented with 15 ?M 6-benzylaminopurine (BAP), were used. The results found that MSB5 most suitable formulation caesia. subsequent experiment, different types cytokinin, BAP, kinetin 2-iP at concentrations 5, 10, 20 ?M, fortified multiplication. multiplication further enhanced by supplementing indole-3-butyric acid (IBA) 1-napthaleneacetic (NAA) 2, 4, 6 8 ?M. showed BAP IBA significantly increased 100% induction, 3.53 shoots/explant, 10.81 cm length, 9.57 leaves, 0.486 g leaves fresh weight 0.039 dry weight. After multiplication, rooting stage carried out altering strength into half full 2.5, 7.5 10 indole-3-acetic (IAA). 5 IAA exhibited highest number roots length roots, 6.13 5.37 cm, respectively. plantlets successfully acclimatized potting cocopeat peatmoss, ratio 1:1 produce survival rate 77.78%. conclusion, established could be useful large-scale raw material production, either conservation extraction.
